Abstract

The characterization of cellular heterogeneity has emerged as a pivotal endeavor in understanding the complexity of biological systems. Single-cell analysis has revolutionized our ability to elucidate the diversity within cell populations, offering insights unattainable through bulk measurements. Microfluidic devices have prominently featured in this domain, enabling high-resolution, sensitive, and multiplexed assays for investigating individual cells with unparalleled precision. This review underscores the pivotal role of microfluidic platforms in facilitating single-cell analysis, discussing their design principles, functionalities, and applications across diverse biological contexts. These devices leverage the precise manipulation of minute fluid volumes, permitting controlled cellular isolation, culture, and analysis. By integrating various functionalities such as cell sorting, lysis, amplification, and detection within confined microenvironments, these systems have enabled the probing of cellular characteristics, including genotypic, phenotypic, and functional attributes at a single-cell level. Furthermore, this review delineates recent advancements and emerging techniques within the realm of microfluidics for single-cell analysis.
